Decoloration and Purification of Polyprenols from Ginkgo Biloba Leaves
Crude non-saponifiable lipid extract contains low purity of polyprenols from Ginkgo Biloba leaves. In order to obtain high purity of ginkgo polyprenols (GP), the decoloration technology of physical adsorption and column chromatography were applied to purify the polyprenols in this paper. The orthogonal experiment was designed to determine the optimum decoloration conditions as followed: dosage of decolorant 12 g, ratio of activated carbon to attapulgite 1:5, temperature 70 ℃, time 20 min. The lipid extract was purified to 49.6% of polyprenols after physical decoloration without loss of polyprenols. Silica gel column chromatography can improve the separation effectiveness of GP and obtain over 90% of GP by gradient elution of ethyl acetate-petroleum ether. This combined purification method, which has such advantages as convenient, fast, economic, high-yield, etc., is very suitable to a large scale production.
