A FACILE METHOD FOR MEASURING INITIAL ADSORPTION OF CELLULASE ON CELLULOSE SUBSTRATE

A facile method based on flow-measurement and UV-visible detection was established for measuring the kinetic adsorption of cellulase on cellulose in the initial stage. A calibration curve relating absorbance at 280 nm and that at 500 nm caused by fine fibers was set up for diminishing the interference of insoluble particles. A rapid and a slow phase can be observed during the steady-state adsorption process and can be described by intraparticle diffusion and pseudo-second-order models, respectively. The method is applicable to pure cellulose and thoroughly washed lignocellulose substrates, spectacularly at low concentration (<2.5%).
