LACCASE PRODUCTION BY PSATHYRELLA CANDOLLEANA IN A STIRRED-TANK FERMENTOR

The laccase production by a white rot fungus, Psathyrella candolleana, was studied in a 5L stirred-tank fermentor. In order to enhance laccase production, Cu2+ and several inducers were applied during fermentation, among which Cu2+ led, by far, to the highest activity (about 23000U/L) in flask culture. The fermentation for laccase production was carried out in 5L fermentor with 1mM CuSO4·5H2O, in which the maximal laccase activity reached to about 9100U/L. The optimum activity of this laccase with ABTS as a substrate was obtained at 45℃ and pH 4.0. It could retain more than 95% of its activity after 40d storage at 4℃ and pH 8.0, which showed that the laccase from P. candolleana had relatively high stability.
