COMPARATIVE THERMAL STABILITY AND ADSORPTION ON VARIOUS BLEACHED FIBER SOURCES OF A RECOMBINANT VOLVARIELLA VOLVACEA ENDOGLUCANASE I (EG1) WITH TS TRUNCATED CATALYTIC CORE (EG1-CD)
Recombinant Volvariella volvacea endoglucanase I (EG1) and its catalytic module (EG1-CD) were purified by two-step chromatography, and the thermal stability and adsorption on bleached fibers were compared. The results indicated that the thermal stability of EG1-CD decreased 17%~28% within the temperature range of 30~60℃ compared with that of EG1, and at lower temperature, a greater gap of thermal stability between EG1 and EG1-CD was found. The binding capacities of EG1 and EG1-CD onto various fiber sources such as bleached wheat straw AP-AQ pulp (WSBAP), hardwood kraft pulp (LBKP), softwood kraft pulp (NBKP) and chemi-thermomechanical pulp(CTMP) were 85%~58% and 45%~25%, respectively. The binding capacities of EG1 and EG1-CD onto various fractions of bleached softwood fiber were found different as well.
