A PRELIMINARY STUDY ON CATALASE ISOZYMES ACTIVITY COMPARISON AND CHARACTER ANALYSIS IN DIP PRODUCTION SYSTEMS
Catalase is an enzyme capable of being produced by all living cells to protect themselves against naturally occurring peroxides. It can rapidly decompose H2O2. This is one of the main reasons to affect the brightness of waste paper pulp or even disable hydrogen peroxide bleaching. In a typical recycling mill, catalase occurs in various sections of the whitewater. This paper studied the catalase activity and analyzed their activity character based on the PAGE (Polyaerylamide Gel Electrophoresis) results and spectrophotometry. The samples were obtained from three sections of the DIP (Deinked Pulp Mill) system. They are pulper section, screen section and disk filter section. The results showed that the catalase from the pulper section had strong activity, the screen section had weaker activity than the pulper section and the disk filter section had the weakest activity. According to results of PAGE, there were 6 enzyme bands in this DIP system and each section had their own characteristic. The pulper section had 4 bands and their activity was the strongest. The screen section had three bands and it was weaker than the pulper section. The disk filter section only contained one catalase band with very dark color which indicated that it had weakest activity. By analyzing this phenomenon, the main reason for this was that survival environments affect the activity of catalase and its kinds. While the survival environment of microorganism has changed, the microorganism is recreated by environment, and therefore the activity and the isozyme of catalase changed.
