CHEMICAL CHARACTERIZATION OF DIFFERENT PARTITIONS DURING THE ISOLATION OF CELLULOLYTIC ENZYME LIGNIN FROM ACACIA CONFUSA MERR. HEARTWOOD

Acacia confusa Merr. is the major hardwood plantation in Taiwan. With outstanding bioactivities, its extractives were used as biomedical purposes; however the extractive-free wood residues were treated as waste. To find an efficient way to utilize the whole biomass and also to alleviate the demand of fossil fuels in Taiwan, these residues are considered a major candidate for biofuel application. Hence, it is essential to understand the characteristics of biomass recalcitrance or lignifications of this tree species before any application for biofuel. In this study, cellulolytic enzyme lignin (CEL) from A. confusa Merr. heartwood was isolated and each different partition was examined during the isolation process. Both lignin contents and structures were characterized. In the current procedure, the total lignin content of CEL is 81.4% and the S/V ratio is 3.0. The purification process seems to partition more S units into dioxane soluble fraction leaving more V unit in residue lignin fraction, causing the S/V rat io of CEL to be higher than that of ball-milled wood (S/V=2.5).
