ON THE DETERMINATION OF LIGNIN CONTENT IN NON-WOOD TISSUES AND PLANTS (2) QUANTITIVE EXAMINATION OF PRE-EXTRACTION TREATMENT
In previous studies, it was strongly suggested that Klason method overestimates the lignin content of non-wood tissues or plants and non-lignin substances are included in Klason residue. In this study, several extractions were applied to ginkgo leaves and zelkova leaves. Klason method and acetyl bromide method overestimate the lignin content when applied to non-wood tissue even after pre-extraction. Klason residue obtained from non-wood tissue contains significant amount of non-lignin substances and preextracted non-wood sample still contains significant amount of non-lignin substances Even by mild pre-extraction, a part of cell wall components (carbohydrate and lignin) are easily removed. Therefore, no pre-extraction method can separate cell-wall components and others selectively and effectively.
