FRACTIONAL ISOLATION AND STRUCTURAL CHARACTERIZATION OF HEMICELLULOSES FROM DENDROCALAMUS MEMBRANACEUS MUNRO

The dewaxed cell walls of Dendrocalamus membranaceus Munro, which were treated with NaC102 and then sequentially with dimethyl sulfoxide, dioxanetriethylamine, 0.5 M, 1 M, 2 M and 3 M KOH, released 9.67, 1.67, 55.15, 12.52, 10.10, and 4.96% of the original hemicelluloses, respectively. The successive treatment together resulted in the dissolution of 94.1% of the original hemicelluloses in cell wall. The chemical and physical characteristics of hemicellulosic fractions were determined by HPAEC, GPC, FT-IR,1 H NMR and 13C NMR spectroscopies. The DMSO-soluble hemicellulosic fractions contained mainly xylose, arabinose, glucose and galactose, of which amount were 86.0, 4.0, 6.2 and 0.5%, respectively; while the dioxane-triethylamine-soluble fractions contained 81.3% xylose, 6.8% arabinose, 2.1% glucose and 2.4% galactose. The four alkalinesoluble fractions were composed of 82.0-90.2% xylose, 4.7-7.5% arabinose, 0.9-3.3% glucose and 0.3-1.5% galacose. Moreover, it was found that the hemicelluloses obtained by dioxane-triethylamine extraction had the relatively lower molecular weight of 17132 g/mol than the hemicellulosic preparations isolated by potassium hydroxide (35430-44712 g/mol). Furthermore, FT-IR,1H and 13C NMR analysis indicated that the alkali-soluble hemicelluloses had a structure composed of the (l-?4) -linked p-D-xylopyranosyl backbone with L-arabinofuranosyl group and 4-0-methyl-a-Dglucuronic acid attached to O-3 and O-2 of xylose in the backbone, respectively.
