STRUCTURAL CHARACTER OF LIGNIN IN FURFURAL RESIDUE AND ITS INFLUENCE ON ACTIVITY OF CELLULASE

Furfural residues, the waste residues obtained from the process of the furfural production with corn cobs, can be used to produce ethanol by the enzymatic hydrolysis and fermentation. However, lignin in the residues will have an influence on the cellulase inevitably during the hydrolysis process. In this study, in order to evaluate the inhibition of native lignin to enzymatic degradation, the milled-woodlignin (MWL) of furfural residual had been separated and characterized by GPC, UV, and 13C NMR spectra. And the enzyme activity of cellulase solution with lignin had also been detected. It was found that the Mw and Mn molecular mass of the furfural residual MWL were 2831 and 2132 g/mol, which were lower than that of corncob. The UV specutu of the MWL of furfural residual exhibited two weak shoulder peaks at around λ280 and 350nm. The signals from 103 to 160 ppm in 13C NMR spectrum of the furfural residual MWL indicate the existence of G, S and H units in the lignin of furfural residual, while the relatively weak signal at 86.2 ppm may imply that the β-O-4 structures are not its main type of connection. The enzymatic activity detection results illustrate that the difference between the enzymatic activity of the original cellulase solutions and that of solution with lignin at first several hours, had a gradual escalation, but it started to decline at 6h (40°C) and 12h (50°C). The difference of 40°C was lower than that of 50°C at corresponding time.So some absorption might be a reversible reaction, which causes desorption of the cellulase from the lignin when the concentration of cellulase is low. The proportion of the absorption and desorption, as well as the stability of the bound between cellulase and lignin, are all effected by temperature.
