Thermal Denaturation Behavior of IFs/IFAP Structures in Merino Wool in a HPDSC under Conditions of Intermediate Regain

Merino wool fibers have been heated to a range of temperatures around the melting point in a High Pressure Different Scanning Calorimeter (HPDSC) under conditions of intermediate regain. The samples were investigated using Raman spectroscopy and polarized optical microscopy. Raman spectroscopy shows in wool samples with 7.3% regain that the helix-coil transition in the crystalline sections of intermediate filaments starts at the onset temperature of melting process. Birefringence experiment shows that after heating to a temperature corresponding to the first peak the fiber are now birefringent on overall of the fiber, and that, furthermore, even after heating to beyond the second peak birefringence is definitely evident. It is suggested that molecular orientation is present in the heated fiber in which the α-helices in the intermediate filament have already melted or denatured.
