REVERSIBLY CROSSLINKED POLY(VINYL ALCOHOL) NANOPARTICLES FOR TRIGGERED RELEASE OF DOXORUBICIN

A novel class of reversibly crosslinked nanoparticles based on poly(vinyl alcohol)-lipoic acid (PVA-LA) conjugates was developed for triggered intracellular release of doxorubicin (DOX). The crosslinked PVA-LA nanoparticles showed excellent stability with less than 20% drug release in 12 h. However, under an intracellular-mimicking reductive condition, rapid and quantitative release of DOX was observed. The intracellular release studies showed that DOX-loaded crosslinked PVA-LA nanoparticles efficiently delivered and released DOX into HeLa cells. DOX loaded crosslinked nanoparticles revealed a similar drug efficacy to the non-crosslinked counterparts.
