CONTROLLED ACID HYDROLYSIS AND ACETYLATION OF GLUCOMANNANS AS DRUG CARRIERS WITH DESIGNED PHARMACOKINETIC BEHAVIORS
Glucomannans with designed molecular weights and acetylation substitution degrees have been prepared by controlled acid hydrolysis and acetylation of Bletillan, obtained from Bletilla Striata. The molecular weights and other structural features were determined by MALS, HPGPC, IR, NMR methods etc. The pharmacokinetic behavior of three samples (BT40, 70 and 110) was studied by intravenous administration in normal rabbits. Plasma elimination half times of these samples were determined to be 5.63, 9.56 and 13.61 hours, respectively. These results indicated that the pharmacokinetic parameters of the glucomannans could be tuned by selection of suitable molecular weights, which is important when these materials are being considered as novel drug delivery carriers.
