DEGRADABLE WATER SOLUBLE HYPERBRANCHED POLYMERS FOR DRUG DELIVERY

Hyperbranched polymers (HBP) with terminal thiol groups and internal hydroxyl groups were prepared from 1,4-dithiothreitol and glyceryl triacrylate. Hydrophilic polymer PEG (550) and cancer drugs (such as camptothecin (CPT) and methotrexate (MTX)) were grafted to the thiol groups and the hydroxyl groups, respectively, forming amphiphilic hyperbranched polymer-drug conjugates, PEG-HPB-CPT, PEG-HBP-MTX and PEG-HBP-S-S-MTX. These conjugates existed as unimicelles in water at low concentration, and self-assembled into multimolecular micelles at high concentration. The MTT experiments showed that the PEG-HBP showed no cytotoxicity and the conjugates had slightly lower cytotosity than the corresponding free drugs, and the subcellular localization of conjugates was observed by confocal fluorescent microscopy, suggesting that the micelles could quickly get into lysosomes in 2 h.
