β-CYCLODEXTRIN-BASED BIODEGRADABLE DENDR1MERS FOR DRUG DELIVERY
Novel polyester dendrimers using β-cyclodextrin (β-CO) as the core and their conjugates with PEG and anticancer drug methotrexate (MTX) were synthesized and characterized by 1H NMR and GPC. The conjugates with the solubilizing group PEG formed nano-micelles in aqueous solution with sizes from 50 to 80 nm. The dendrimer-MTX conjugates showed effective activity against breast cancer (BC) in vitro with an IC50 of 0.068 ng/mL, and the subcellular localization of the conjugates was observed by confocal fluorescent microscopy, indicating that the conjugates mainly localized in lysosomes.
