PHOTO-CROSSLINKED BIODEGRADABLE MICELLES FOR PACLITAXEL RELEASE

Crosslinked biodegradable micelles were developed based on poly (ethylene glycol)-poly (acryloyl carbonate)-polylactide (PEG-PAC-PLA) triblock copolymer. The facile combination of PEG-PAC-PLA and folate-PEG-PLA yielded stable micelles with varying folate densities. The crosslinked micelles had sizes ranging from 85 to 140 run with a low PDI of 0.05-0.14. MTT assay using KB cells revealed that these crosslinked micelles were not cytotoxic. These micelles showed high loading efficiencies for paclitaxel (PTX) and minimal drug release upon dilution. The enhanced drug stability combined with active targeting may effectively improve tumor-targetability of biodegradable micelles.
