Optimization of Extract Conditions of Protein from Wheat Bran by Enzymatic Process Using Responses Surface Methodology

Response Surface Methodology (RSM) consists of a series of mathematic and statistical methods, and can be used to determine the effect of factors and their interactions to dependent variables during processing. Established mathematical model can be used to express the relationship between response values and factors exactly and to predict experimental value under different process conditions. On the basis of single factor test, RSM is used to optimize the enzymatic extraction conditions of protein from wheat bran. The optimum conditions are: 57.9°C, pH8.4, E/S 5.8%, substrate concentration 9.3 %, time 2h. The results of verification of the model and analysis of variance (ANOVA) for the model show that the model is significant (p<0.01) and can be used to predict the protein yields under different extract conditions.
