Expression of hepatitis C virus F protein leads transfected cell to transformation
In this study, we aim to investigate the role of HCV F on the cellular transformation. HCV F gene was amplified using PCR and cloned into the eukaryotic expression plastnid pcDNA3. 0. NIH3T3 cells were transfected with pcDNA3-F plasmid using Lipofectamine 2000 transfection reagent. Anchorage-independent growth in soft agar was then carried out. Transfected cell lines were obtained from experimental transfections, which showed anchorage-independent growth in soft agar. In conclusion, overexpression of the F protein resulted in an increased proliferation rate in NIH3T3 cells, the core protein expression also did induce cellular transformation.
