Expression and Bioactivity of Recombinant Human Cytoglobin
To investigate the function and pharmaceutical application of cytoglobin. Cytoglobin cDNA was cloned from cDNA library of HepG2 cell by RT-PCR and then subcloned into expression vector pET28a. The recombinant expression vector was transformed into the E. coli BL21, the genetic engineering strain pET28a-rhCYGB/BL21 was constructed, the strain was fermentated in large scale (5L). The expression of rhCYGB was induced by IPTG at concentration of 0. 5 mM. The purified rhCYGB (95%) was obtained through several purification steps. The in vitro and in vivo experiments demonstrated that recombinant human CYGB has antioxidant activity and could be a potential biotech drug for treatment of liver fibrosis.
