A molecular linkage map of sweetpotato based on SRAP markers

SRAP markers were applied to construct molecular genetic linkage maps of sweetpotato with 240 individuals of the F, population derived from a cross between sweetpotato Luoxushu 8 and Zheng-shu20. A total of 1 337 polymorphic molecular markers were generated with 344 SRAP primer combinations. There were 873 markers obtained in Luoxushu 8,which included 507 Simplex,212 Duplex,31 Triplex, and 123 Double-simplex intercross markers; 587 markers were obtained in Zhengshu 20, which included 356 Simplex, 104 Duplex,4 Triplex, 123 Double-simplex intercross and 85 distorted markers. U-sing the software of JoinMap 3. 0 and the double pseudo testcross strategy ,a molecular linkage map containing 90 linkage groups was constructed with 514 SRAP markers in Luoxushu 8,and the map coveed 6 720.03 cM with an average marker interval of 10.50 cM. In Zhengshu 20,the molecular linkage map containing 79 linkage groups was constructed using 384 SRAP markers, and the map covered 4 768.62 cM with an average marker interval of 12.42 cM. 11 and 12 homologous groups were identified by 104 and 28 multiple-dose markers in the Luoxushu 8 and Zhengshu 20 maps,respectively. There were 29 Double-simplex intercross markers present in paternal and maternal linkage maps,which allowed the identification of the 45 corresponding linkage groups between both parents.
