Assembly, functional annotation and classification of 22 726 ESTs from sweet potato I pomoea batatas (L.) Lam.
Expressed Sequence Tag (EST) has high functional information content,and often corresponds to genes with known or predicted functions. In this paper,we retrieved all available EST sequences for sweet potato (Ipomoea batatas) from NCBI (National Center for Biotechnology Information) GenBank. After trimming and discarding low quality and contaminated sequences, the resulting high-quality ESTs were used for further analysis. Sequence clustering resulted in 3 407 contigs and 4 856 singletons. Based on sequence similarity search,total 5 657 unique ESTs could find hit in UniProt database with E value cutoff of less than 10~5. The other 2 606 ESTs with no hit were also identified. In order to further characterize and categorize biological and molecular functions of these unique EST sequences,Gene Ontology (GO) was used for classification annotation. Of these sequences,3 082,1 455,and 2 313 were assigned to biological processes,molecular functions,and cellular components,respectively. These results provided a convenient resource for next research,such as SSR marker development,SNP discover-y,functional gene identification and oligonucleotide design for microarray analysis.
