AFLP analysis among different ploidy levels of I pomoea species
Cultivar sweetpotato is a hexaploid species,of which the exact proceeding of origin and domestication of polyploidization has not been well defined. A diploid progenitor,a naturally crossed tetra-ploid,and three artificially synthesized tetraploid were used as materials to study the relationship between ploidy variations and numbers of DNA bands,to explore the phylogentic relationship of three new artificially sythesized interspicific hybrids with sweetpotato cultivar. The results show that the number of amplified AFLP bands is increasing accompanied with the increase of ploidy levels. The similarity coefficients among all pairs of six Ipomoea species varied from 0.54 to 0.90,all showing above 50% genetic distance. The similarity coefficients between Xushu 18 and other 5 Ipomoea species are different and the order from high to low is as the following:H67-1 ,H10-1 ,H14-1, I. trifida and/. tabascana. It appears that the three new artificial hybrids are close relatives of Xushu18.
