Design of the Fluorescence Detector Device Based on Integrating Sphere

The fluorescent detection device was designed based on LED and integrating sphere. System used monochromatic LED as the light source, which through the interference filter and integrating sphere of uniform radiation to reduce the influence of light on the sample. When the light source went through the PMT and follow-up circuit, the real-time PCR cycle fluorescence intensity was gotten, finally. This design could effectively reduce the volume of fluorescence detection device and improved the costeffective of instrument. The experiment was compared the results between the device and iCycler-iQ5 PCR instrument which excitation light source dose not have homogenization. The instrumental maximum deviations are 9.4% and 26.0%. And the deviation of iCycler-iQS PCR instrument is 13.9%. Its obvious that the device have higher accuracy, better stability and reproducibility.
