Enhanced Production of Recombinant Thermobifida fusca Cutinase by TIR Degeneracy Mutagenesis

Introduction As the threshold of an intracellular translation process, the Translational Initial Region (TIR) plays a constant key role in determining the whole efficiency of protein translation. Tiny alternations in TIR which extends from immediately from upstream of the Shine-Dalgarno sequence to approximately twenty nucleotides downstream of the initiation codon were reported to change the subsequent downstream translation process (1). Such property provides not only the importance of TIR to the global translation efficiency, but also a powerful potential tool for optimization of a specific recombinant protein. In present study, degeneracy mutagenesis was applied in pelB which is the most widely used signal peptide in E.coli for extracellular production of recombinant protein to enhance the expression level of cutinase from Thermobifida fusca.
