Comparison of 12 different melting temperature calculation methods for short oligonucleotides
Melting temperature (Tm) of oligonucleotide is the key factor for the success of several popular molecular techniques, such as quantitative polymerase chain reaction (PCR), multiplex-PCR and DNA microarrays of short sequences with fixed length. Although there are more than 20 methods or variants available for prediction of the Tm value, the difference of predicting accuracy is quite large and brings much trouble when designing a suitable PCR primer or probes for DNA microarray. Therefore, its important to compare these methods and find out which is the most accurate predicting method.
