EFFECT OF THE EXTRACT OF BRUCEA JAVANICA (YA DANZI) ON NO/CGMP IN MURINE SPLENOCYTES
In order to investigate the effect of the extract of Brucea javanica (Ya Dan Zi 鸦胆子提取物) on the signal transducing system of NO-cGMP in murine splenocytes, the mice were taken as the experimental object, the spleen lymphocyte was separated and divided into four groups according to the medicines density: 0 μ g/mL (control), 80 μ g/mL, 120 μ g/mL and 240 μ g/mL, cultivated together with the medicine, the cells and the supernate were collected in 4 h, 8 h, 12 h, 24 h respectively. The NO and cGMP in splenocytes and the supernate were detected by the means of Griess and radioimmunoassay respectively. Results: the content of NO was in dose-dependant and time-dependant during the period of culture of the Brucea javanica. At the concentration of 80μg/mL and 120μg/mL, the content of cGMP reached maximum at 8th hour. At the concentration of 240μg/mL, the content of cGMP was decreased with the prolongation of culture time. By analyzing change of NO and cGMP, there was a certain relationship between them. It was concluded that the Brucea javanica played the important role in regulating signal transduction system of NO/cGMP.
