Citrus and Colon Cancer Prevention:Current Research and Future Prospects

Colorectal carcinoma is one of the leading causes of death throughout the world and prevalence of this disease is believed to be related with dietary habits. Based on numerous evidences using both in vitro and in vivo studies, natural bioactive compounds from fruits and vegetables are gaining significance as colon cancer preventives. Our recent research suggests that certain citrus bioactive compounds and grapefruit extract are capable of preventing colon cancer effectively. In vitro studies conducted in human colon carcinoma (HT-29)cells have demonstrated that citrus limonoids can effectively inhibit growth of cells. In another study phytosterol and aliphatic esters from citrus have also shown inhibitory effects on these cells and in both the cases compounds were found to be non toxic to non-cancerous cells COS-1 cells, which indicate possibility of ideal components and/or fruit for colon cancer prevention. It seems that these compounds are known to inhibit through induction of caspase 3/7 activity leading to apoptosis. Recently, we have also found that both limonoid aglycone and their respective glucosides are capable of inhibiting colon cancer (SW-480) cells through intrinsic apoptotic pathway. Additionally, our previous animal study has revealed that, freeze dried grapefruit juice powder and its bioactive compounds such as naringin and limonin are capable of inhibiting aberrant crypts through suppression of cyclooxygenase-2 and inducible nitric oxide synthase (iNOS) in azoxymethane treated animals. These findings suggest that citrus and their bioactive compounds can serve as promising source for prevention of colon cancer.
