Studies on Plant Morphology and Genetic Diversity of Natural Tetraploid and Diploid of Citrus sinensis cv. Hongjiangcheng

Hongjiangcheng (C. sinensis) is an elite sweet orange cultivar in Guangdong province of China. But it is seedy with about 20 seeds per fruit. Tetraploids can be used to seedless triploid breeding. In order to elucidate genetic background and plants character of our tetraploid plants, comparation of natural tetraploids of Hongjiangcheng with correspongding diploid was undertaked on plant morphology and genetic diversity. Genomic DNA variation was analyzed by GISH (Genomic in situ hybridization)and SSR. Compared with diploid, the characteristic of tetraploid plants of Hongjiangcheng was poor growth, small tree crown, 0.8 -0.9 times height and circumference of the trunk, short but dense vernal branches, thick and long thorns. Leaves of tetraploids were broader, obvious thicker and darker in color than leaves of diploids. Using genomic DNA of diploid for probe, natural tetraploids was proved autotetraploid by GISH. SSR bands analysis showed that eight pairs among twentyone pairs of SSR primers displayed different bands, which suggested that there was DNA variation on natural autotetraploid with corresponding diploid. So, there were obvious diffences on plant morphology and DNA variation between the tetraploid and diploid of Hongjiangcheng.
