cDNA-AFLP Identifies Differently Expressed Transcripts of a Novel Late-ripening Citrus sinensis Mutant Potentially elated to Fruit Ripening
Fengjiewancheng (FJWC) is a late-ripening bud sport of Fengjie72-1 navel orange (Citrus sinensis Osbeck). In the present research, using cDNA-amplified fragment length polymorphism, we compared the transcript difference between fruits of the two varieties during ripening. As a result, about 60 000 clear transcript-derived fragments (TDFs) were generated with band sizes ranging from 50 to 500bp, of which 127 TDFs were down regulated (88.2%) and 17 were up-regulated (11.8%) in FJWC while compared with FJ72-1. Blast annotation indicated that 39 uniTDFs, which mainly located in mitochondria (26%)and membrane system (44%), were assigned with putative function in signal transduction, control of gene expression and carbohydrate biosynthesis, involving in extensive biological processes such as response to stimulus (12%), transport (12%), cellular metabolic process (12%), post-translational protein modification (12%), micromolecule biosynthesis (12%), transcription (10%), cellular localization (10%), phosphate metabolic process (10%)and developmental process (10% ). In addition, some TDFs showed relatively high identities with known functional genes and had differential expression patterns in fruit pulp or peel between FJWC and FJ72-1 during fruit ripening, implying a potential role in the formation of the late-ripening phenotype.
