Application of SSR Molecular Marker in the Identification of Citrus Varieties
Simple sequence repeat (SSR) marker was used to study the relationships of the sixty three of citrus collections. A total of fifteen SSR pairs of primer were screened out of 22 primer pairs. A total of 277 amplified bands were detected, among which 267 polymorphic bands with averages of 6~36 bands produced by each pair. The results showed that all the cultivars among the collections shared more than 0.7 of genetic similarity, of which these were 0.964, 0.988 and 0. 913 of genetic similarity coefficient for the different varieties ofsweet orange (Citrus sinensis) , among different varieties of satsuma (C. unshiu) and among different varieties ofponkan (C. reticulata) , respectively. Nevertheless, it was existed wide variation range of genetic similarity among other citrus hybrids. The obvious genetic divergence was observed among species or cultivars of Fortunella crassifolia Swingle, C. limon Burm. , Citrus aurantifolia Swingle, C. grandis Osb. , C. reticulata Blanco and C. sinensis Osb. , respectively. It could be differentiated from each other except Seike navel and Shirayanagi navel, Gailiangcheng and Tarocco orange Xinxi , Ponkan 56-17 and Yanxiwanlu ponkan.
