Adventitious Shoot Regeneration from Five Commercial Citrus Cultivars Using a Range of Explant Types

The parameters for optimum regeneration of five commercial citrus cultivars were tested using a range of explants. Different optimum medium for shoot regeneration of the citrus cultivars Sihuitianju (Citrus retkulata Blanco.), Shatangju (C. reticulata Blanco.), Ponkan (C. reticulata Blanco.), Gonggan (natural hybrid cultivars of C. sinensis X C. reticulata Blanco.)and Anliucheng (C. sinensis Osbeck) were achieved using epicotyl segments, hypocotyl segments and cotyledon as explants respectively. The optimum medium for shoot regeneration of Sihuitianju was MT medium supplemented with 5 mg/L 6-BA and 0.5 mg/L IBA; the optimum medium for Shatangju was MT medium supplemented with 2. 5 mg/L 6-BA and 0. 5 mg/L NAA; the optimum medium for f Ponkan was MT medium supplemented with 0.5 mg/L NAA; the optimum medium for Gonggan was MT medium supplemented with 7. 5 mg/L 6-BA and the optimum medium for Anliucheng was MT medium supplemented with 2.5 mg/L 6-BA and 0.5 mg/L IBA. Regeneration from epicotyl proved to a promising explant source while cotyledon proved to be difficult from all explant materials. This system thus provides a convenient method that could be adopted for commercial citrus in vitro propagation or genetic transformation of commercial citrus varieties.
