Variability of Fusarium spp. in Citrus Rhizosphere

Citrus dry root rot disease is directly associated to Fusarium solani (Mart.)Sacc, and indirectly related to other biotic and abiotic factors. In this work, the identification and grouping of Fusarium spp. isolated in the citrus rhizosphere were investigated. Soil, root and wood samples were collected from citrus nurseries and groves in different Mediterranean locations. Fusarium spp. were isolated on a selective medium and morphologically characterized on carnation-leaf agar (CLA) and low-nutrient agar (LNA). Molecular identification was carried out by DNA extraction, PCR amplification of β-tubulin and α-Elongation Factor loci using specific primers, sequencing the PCR products and searching for the nucleotide sequence identity in NCBI database. F. solani, F. oxysporum and F. proliferatum were isolated from soil samples and groves, whereas only F. solani and F. oxysporum were isolated from wood and feeder roots. Neighbour phylogenetic trees based on nucleotide sequences of the fungal isolates were obtained; genetic groups were heterogeneously assembled in two main groups which consisted of Fusarium spp. sources with different origin (soil, root or wood). Genetic fungal species patterns showed no correlation with the isolate geographical origin.
