Isolation and Purification of Anthocyanins from Blood Oranges by Column Chromatography
Blood orange anthocyanins were purified by a NKA-9 resin column and a Toyopearl TSK HW-40S column chromatography. The best separation was obtained using a mobile phase of 35% methanol with 2% formic acid at a flow-rate of 0. 6ml/min. Three major anthocyanins of blood orange were isolated. They were identified as cyanidin-3-glucoside, cyanidin-3-(6malonyl) glucoside and cyanidin 3-glucoside-derived pyranoanthocyanins by HPLC and HPLC-MS. Furthermore, cyanidin-3-(3-malonyl) glucoside was identified for the first time in blood orange juice. The method of purification in our study is easy to use and obtain pure anthocyanins. It is helpful for the further study, such as research of degradation and physiological activity.
