Preparation of Immobilized Protease on the Magnetic Chitosan Microspheres and Its Application on Wool Treatment

Immobilized protease on the magnetic chitosan microspheres was prepared with glutaraldehyde as cross-linking agent. The effects of immobilization conditions, the stabilities and the recovery properties were determined. And then the immobilized protease was applied on the wool fabric and the finishing results were tested. When pH was 7.5, the temperature was 40℃, the concentrations of EDC and glutaraldehyde were respectively 0. 25% (W/W) and 2. 0% (W/W), the concentration of enzyme was 0.4% (V/V), the relative activity of the immobilized protease reached 66%. Compared with native protease, the pH, thermal and storage stabilities of the immobilized protease were greatly improved. In addition, the immobilized protease had desirable reusable and was used for 30 times with about 81% ratio of activity recovery. The prepared immobilized protease was used for treatment of wool fabric, the area shrinkage rate was 7.1%, the weight loss percentage was 6.4% and the strength loss percentage was 8.2%. These results confirm that when treating wool fabrics with immobilized protease, by the proper treatment conditions, the good levels of shrink resistance without considerably damaging the fabric can be achieved.
