Improved Cellular Response on the Multiwalled Carbon Nanotube-lncorporated Electrospun Chitosan/Poly (vinyl alcohol) Nanofibers

We report the fabrication of multiwalled carbon nanotube (MWCNT)-incorporated electrospun chitosan (CS)/poly (vinyl alcohol) (PVA) nanofibers with high water stability for tissue engineering applications. In this study, electrospinning parameters (e. g., voltage, collection distance, and polymer concentration) and crosslinking conditions were optimized to form smooth and uniform CS/PVA and MWCNT/CS/PVA nanofibers with high water stability. The morphology, structure, and mechanical properties of the formed electrospun fibrous mats were characterized using scanning electron microscopy (SEM), Fourier transform infrared spectroscopy, and mechanical testing. We show that the incorporation of MWCNTs does not appreciably impact the morphology of the CS/PVA nanofibers, importantly the tensile strength and elongation characteristics of the nanofibers are significantly enhanced when comparing to those without the incorporation of MWCNTs. These scaffolds with small diameter (around 150 nm) and high porosity can well imitate the natural extracellular matrix. In vitro MTT 3-(4, 5-dimethylthiazol-2- yl)-2,5- diphenyl tetrazolium bromide assay and SEM morphology studies show that the incorporation of MWCNTs into the nanofibers mats significantly promotes cell attachment and proliferation. Given the unique fibrous structure of the nanofibers and the excellent mechanical properties, the MWCNTincorporated nanofibers should be applicable in the fields of tissue engineering and regenerative medicine.
