DEVELOPMENT OF DISTRIBUTIONS OF RELATIVE POTENCY ESTIMATES TO QUANTITATIVELY ASSESS UNCERTAINTY INHERENT IN THE TEFS FOR DIOXIN-LIKE COMPOUNDS: A PROPOSED CONSENSUS-BASED WEIGHTING FRAMEWORK

The toxic equivalency factor (TEF) methodology currently serves as the basis for estimating potential health risks that could theoretically occur as a result of exposures to mixtures of dioxin-like compounds. The TEF values represent single point estimates recommended by the World Health Organization (WHO). One limitation of these point estimate TEFs is that it is not possible to capture the uncertainty inherent in each of these values. Being able to quantify this uncertainty is important given that the relative estimates of potency (REP) values underlying each of the WHO TEFs span several orders of magnitude. Some investigators have recommended the use of the underlying distributions of REP values to develop probabilistic estimates of risk as a means of quantifying the inherent uncertainty. A key component of such an approach involves weighting the REP values in the distributions for each congener to place greater emphasis on those REP values that are believed to be better suited for health risk assessment purposes. As such, the goal of this study was to develop a consensus-based weighting framework that would account for differences in study quality and relevance. The consensus-based weighting framework that has been developed incorporates several different objective measures of study quality and relevance including study type, pharmacokinetic considerations, endpoint type, and REP derivation method and quality. Multiple iterations and numerical scales were assessed in the process of developing the consensus-based weighting framework proposed herein.
