Bifunctional Magnetic SERS Probes based on Thrombin Aptamer and its Application in Thrombin Detection

Aptamer has been widely used in specific molecular recognitions from inorganic and organic species to proteins and cells 1. Until now, many methods based on aptamer to recognize specific targets have been developed such as surface plasmon resonance (SPR) 2, atomic force microscopy (AFM) 3, fluorescence 4, colorimetry 5, electrochemistry 6, and so on. Surface enhanced Raman scattering (SERS), which is a powerful tool in biologic analysis for its high sensitivity and little disturbance from water, is also used to fabricate new sensors based on aptamer to detect protein with quite low detection limit and high selectivity 7. However, for the existed aptasensors, nonspecific absorption is a problem which would influence the sensitivity and reliability. To reduce this influence, complex processes, such as repeated wash or blocking with special molecule, are usually needed. Herein we developed bifunctional magnetic SERS probes based on aptamer which could simplify the process and reduce the influence of nonspecific absorption. Meanwhile the detection limit is comparable to other methods.
