LIPID METABOLITES PRODUCED BY SELECTIVE WHITE ROT FUNGI

A selective white rot fungus, Ceriporiopsis subvermispora is able to delignify wood without penetration of extracellular enzymes into wood cell wall regions. We reported that the fungus produced manganese peroxidase (MnP) and free fatty acids to drive MnP-dependent lipid peroxidation at an early stage of wood decay within a month. After prolonged cultivation, the fungus produced a series of alk(en) ylitaconic acids (ceriporic acid A, B, C) during the wood decay. One of the metabolites, l-nonadecene-2,3dicarboxylic acid (ceriporic acid B) inhibited iron redox cycle, thereby suppressed OH production and depolymerization of cellulose by the Fenton reaction. Thus, lipid-related metabolites play a central role in the selective ligninolysis. In the present study, profiles of the lipid-related metabolites including ceriporic acids and free fatty acids on different liquid culture media, glucose-yeast extract (GY), glucose-peptone-yeast extract (PGY) and yeast-maltpeptone-glucose (YMPG) media were analyzed. The highest production of ceriporic acids was observed on the GY medium. In the series of ceriporic acids, production of ceriporic acid C was the highest in all the culture media examined. On cultivation in GY medium, the fungus produced palmitic (16:0), stearic (18:0), oleic (18:1 cis-Δ9), vaccenic (18:1 cis-Δ11), and linoleic (18:2 cis-Δ9,A12) acids as major saturated and unsaturated fatty acids. Among them, the most abundant was linoleic acid, which consists of 82% of the total fatty acids after 7 days. Production of linoleic acid increased during the first 7days, while production of ceriporic acids continued for 30 days on the GY medium. Thus it was found that the fungus differentially regulates production of the lipid-related metabolites in the liquid cultures.
