In Situ Imaging of Breast Cancer Cells using Green Semiconductor Quantum Dots

We report the development of early stage in situ imaging of breast cancer cells (SK-BR3) using green quantum dots (QDs) as fluorescent signal generator. The QDs based imaging of breast cancer cells involved anti- HER2/neu antibody for labeling the over expressed HER2 on the surface of SK-BR3 cells. The complete assay involved SK-BR3+Ab-biotin+streptavidin~QDs. The SK-BR3 cells were grown in culture plated, exposed to the biotin labeled antibodies and later exposed to streptavidin labeled QDs to utilize the strong and stable biotinstreptavidin interaction. Fluorescent images of the complete assay for SK-BR3 cells were evaluated under a microscope with a UV light source. The preliminary results showed the SK-BR3 in the complete assay as fluorescent cells that had brighter signals compared with those labeled with organic dye using similar parameters and the same number of cells. Our future directions involve optimization of the various parameters of the assay, establishment of dynamic range of concentrations detected as well as limits of detection, and multiplex detection of different types cells and/or biomarkers of cancer.
