Intelligent Surfaces for Cell Sheet Tissue Engineering

Our ideal therapy with tissue engineering is achieved by cell sheet manipulations engineering that offers the two following key features. One feature that allows noninvasive harvest of cultured cells as cell sheet is realized by the development of the temperature-responsive culture dishes. The surface of the dishes is hydrophobic at 37℃, but it reversibly changes to hydrophilic below 32℃. Various cell lines adhere, spread, and proliferate on the surface in the same manner as the cell lines do on commercially available regular tissue culture dishes. Only by reducing the temperature, cells spontaneously detach from the surface without the need for trypsin or EDTA. As a result, confluent cells are recovered as a single contiguous monolayer sheet with deposited extracellular matrix intact. The other feature is that the harvested viable cell sheets can be transferred to other biological surfaces (2D cell sheet manipulation) because the extracellular matrix on the basal side of cell sheets is biologically adhesive. Thus, tissue regeneration with cell sheet tissue engineering can be accomplished by transplantation of single cell sheets, as with skin, cornea, and periodontal ligaments. We have already initiated human clinical study of cell sheet engineering therapy using oral mucosal cell sheet for cornea epithelium deficient disease and for esophageal epithelial cancer after endoscopic submucosal dissension surgery. We also have succeeded in treating cardio-myopathy using myoblast cell sheet.
