The Research of Penicillin Sulfozide Crystallization

Penicillin sulfoxide is the precursor of 7-ADCA which is one of the important nucleuses of cephalosporin antibiotic. The quality of penicillin sulfoxide is crucial to the product of 7-ADCA. The white crystal was changed into yellow powder with the purity reduced. Crystal structure and constitute take great efforts on the degradation rate. So some operation conditions were studied in the paper. Solubility of penicillin sulfoxide in water was experimentally determined. The solubility of penicillin sulfoxide is affected by temperature and pH evidently. It increases with the increase of pH and temperature. The controlling of pH and temperature is the key operation of penicillin sulfoxide crystallization. There is a balance between solution and surfaces, the surfaces of the crystal become coarser as the temperature increase. The crystal size distribution changed with the increase of feed rate, the main size of penicillin sulfoxide become smaller with either big or small feed rate. The main size of crystal of penicillin sulfoxide is 159.92 μm with the feed rate of 0.45 ml/min, contrast with 140.21 μm when the feed rate is 0.35 ml/min. With the increase of agitating time, the plantlike crystals were changed into clava. But the crystals which agitated for 2 h were similar with that agitated for 3 h. It’s needlessly choose too long time for the process. The concentrations also take efforts on the crystal size distribution. The C.V decrease first and then increase with the enhanced of concentration. When the concentration is 45 mg/ml, the C.V of penicillin sulfoxide reaches the smallest.
