Study on the Process of Reactive Crystallization of Clopidogrel Bisulfate

In this article, the phenomena of polymorphic transformation is revealed in the process of preparation of clopidogrel, which was achieved by reactive crystallization with acetone as the solvent In the process, we sampled in the regular time, and then identified with X-ray powder diffraction analysis. The result showed a transformation from polymorph I to polymorph II, as is reported form II is more stable than form I in thermodynamics. For further study, we observed the sample by SEM, and found that form I is plate-like while form II is club-shaped, both coalesced dramatically. In addition, much work was done in order to control the process of the transform, and the discipline was disclosed that the occurrence was delayed with temperature decrease and concentration increase. By further research, we explained this discipline, which provided a theoretical foundation for the production of this drug.
