A Novel Approach to Investigate the Primary Homogeneous Nucleation Kinetics of Anti-solvent Crystallization

A novel approach is presented to investigate the primary homogeneous nucleation kinetics for the anti-solvent crystallization of Deflazacort in binary n-hexane+ethyl acetate, n-hexane+acetone solvent systems at 303.15 K using Laser Monitoring Technique. This method is based on the classical theory of 3D nucleation and the assumed power-law relation between the particle number density (N/V) and the opacity detected by Laser Monitoring Technique. The transmittance of laser beams is correlated with time to obtain a series of curves for both systems using the Boltzmann equation. The primary homogeneous nucleation kinetics is interpreted by the relation between the calculated slope of these curves at constant transmittance and the initial supersaturation ratio. The crystal-solution interfacial tension γ and the surface entropy factor f can be calculated from the kinetic parameters.
