Solid Dispersion Facilitating the Osmotic Pump Delivery of Water-insoluble Drug
Monolithic osmotic pump tablet (MOPT) is benefited by the simplified preparation process. Drug release from the MOPT depends on the drug water-solubility and dispersion rate in the polymeric matrix of tablet. As the low solubility of drug is one of the most important factors affecting the drug release process, many methods have been proposed to enhance the drug solubility. It was repoeted that inclusion complex was applied to promote drug solubility and stability in MOTS for water-insoluble drug. However, not all drugs could form inclusion complex and it may be more suitable to delivery drugs in suspension. In the present study, the MOPT composing drug solid-dispersion, which was widely used to increase the solubility, dissolution rate and bioavailability of water-insoluble drugs, was developed to facilitate the constant drug release from the MOPT.
