In Vivo and In Vitro Structural Variations of The Injectable Chitosan-based Hydrogel Drug Delivery System

Hydrogel is three-dimensional, cross-linked network of water-soluble polymers. For the reason of good biocompatibility and flexibility in application, hydrogels are commonly used in clinical practice and experimental medicine for a wide range of applications. Thermosensitive hydrogels such as chitosan-β-glycerophosphate (CS-GP) have been considered as novel drug delivery matrices and attracted more and more attentions recent years. They can be subcutaneously injected at room temperature, form a gel drug depot in-situ and control drug release for a long term. However, the subcutaneous release environment, which might influence the gels’ conformations and the drug release, was found quite different from the release environment in vitro. In this study, the structural variations of CS-GP hydrogel were investigated in vivo and in vitro, respectively, in order to achieve a better prediction of drug release.
