Relation between Cellulases Activity and Strains in a Lignocellulose-degradation Bacterial Community
In order to find out the dynamic changes of four kinds of cellulases and hemi cellulase in fermentation progress, the standard cellulase activity assays published by the International Union of Pure and Applied Chemistry (IUPAC) was used. The results showed that enzyme activities were all one signal peak curve, endoglucanases, exoglucanases, a -glucosidases and the total cellulase activity reached the maximum on the day 8, the value were 4.48, 15.83, 25.78 and 7.51 U/mL , respectively. While compared with cellulase, the maximum of hemi cellulase appeared earlier , it reached to the maximum 280.9 U/mL on the day 5. For find out the relation between enzyme activity and strains in bacterial community, a technology of polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) was used to profile the stru ctured dynamic changes of microbial communities in fermentation process. The result showed that the strain of Bacteria and Paenibacillus could promote excrete xylanase. And the strain of Clostridium could excrete cellulase and xylanase, simultaneity.
