A Rapid and Convenient Estimation of Bacterial Intracellular Polyphosphate Content by Using Photometric Method

Estimation of intracellular phosphorus content especially polyphosphate content by photometric method was assessed. Basic dyes such as methylene blue and toluidine blue was utilized to stain bacterial cell suspension. In order to evaluate staining intensity, DIFsp, which was defined as maximum absorbance of difference spectrum between stained- and non-stained cell suspensions followed by corrected at 1000 nm, was increased along with the increase of total phosphorus content in polyphosphate accumulating mutant Escherichia coli strain MT29 (correlation coefficient: 0.97). DIFsp also showed a good correlation with polyphosphate content in E. coli mutant strain. The sensitivity of DIFsp (methylene blue) was 1.6 fold greater than that of DIFsp (toluidine blue). This rapid and convenient method for estimation of intracellular polyphosphate content was applicable to screening polyphosphate accumulating bacteria to aid elucidation of enhanced biological phosphorus removal.
