Copper Accumulation and Eco-physiological and Biochemical Responses of Androgenic Carrot Cultures to Copper Stress

Carrot easily accumulates toxins, so it is very important to use in vitro cultures in selecting and eliminating the cultivars which accumulate excessive amounts of metals. Long term influence of Cu2+ on regeneration and growth of carrot genotype Kazan obtained from androgenic embryos was analyzed. Copper was added to the medium at concentrations: 0.1 μM (control), 1 μM, 10 μM and 100 μM. The cultivation at 10 μM Cu2+ concentration resulted in stimulation of growth during 4 and 9 weeks, even if TBARS level also increased. At the same time a significant increase in phenolic compounds absorbing UV light at 330 nm was observed. It seems that flavonoids rather than free proline may be involved in protective mechanisms against Cu2+ stress during short term regeneration of carrot androgenetic embryos. The highest Cu2+ concentration (100 μM) enhanced lipid peroxidation, and although after 9 weeks of cultivation the proline level significantly increased, the Cu accumulation in tissues was almost doubled in comparison with the control and growth limitation was caused. Rosettes growing at low concentrations of Cu2+ (0.1 and 1 μM) accumulated less Cu over time, which was parallel to their high organogenic ability and could suggest their acclimation to stress conditions.
