Determination of Microcystins-LR in Aquatic Products Stocked in the Lake Taihu Using High Performance Liquid Chromatography
The presence of toxic cyanobacterial blooms has caused severe problems for aquatic animals. Monitoring of microcystin LR (MC-LR) in fish and other aquatic animals is important for evaluating the potential risk for human consumption. A solid phase extraction (SPE) with liquid chromatography ultraviolet spectrometry (LC/UV and LC/MS) method has been developed to identify and quantify MC-LR in aquatic animals. The extracted procedure was determined by comparing different solvents, homogenization types and time, respectively. The study showed decreasing of recovery via blowing to dryness by nitrogen stream. The recovery, increasing with added quantities of MC-LR (0.17 μg·g-3.33 μg·g-1 DW), was from 70% to 80% with high consistency (RSDs<5%). The limit of detection was 0.067 μg·g-1 DW; this analytical method presented a simple and rapid procedure for determining MC-LR, and could apply to most of aquatic animals. Among the aquatic animal samples collected from the Lake Taihu in 2007 and 2008, 25% were detected containing MC-LR, indicating that aquatic animals were risky for consumption in Lake Taihu. The results of this study provide the scientific base for establishing the tolerance limit of microcystin in aquaculture products.
