Microbial Ecology in Acid Mine Water Samples and IscA in Acidithiobacillus Ferroozidans
Microbial ecology and community compositions in the acid mine water samples from Dongxiang Copper Mine (in Jiangxi province), Yinshan Lead-zinc Mine (in Jiangxi province), Yunfu Sulfide mine (in Guang-dong province), Zhongtiaoshan Copper Mine (in Shanxi province) and Daye Copper Mine (in Hubei province) were studied by 16S rDNA cloning and Restriction Fragment Length Polymorphism (RFLP) methods. Bacteria detected from the samples were fell into six putative phylogenetic divisions, which were α-Proteobacteria, β-Proteobacteria, γ-Proteobacteria, Firmicutes, Actinobacteria and Nitrospira. Archaea was only detected in the acid mineral samples from Dongxiang Copper Mine, Yinshan Lead-zinc Mine and Daye Copper Mine. The adsorption of microbes, which were from Yunfu Sulfide mine, on pyrite specimen surface was studied by SEM (scanning electrode microscopy) and FISH (fluorescence in situ hybridization). The results showed A. ferrooxidans (Acidithiobacillus ferrooxidans) intended to be absorbed on the pyrite surface. And A. ferrooxidans would set themselves as nucleus adsorbing ferroalumen precipitation, and Leptospirillum species adsorbed on the pyrite surface were likely to accumulate together and keep surface smooth. In addition, the key protein in Fe-S cluster assembly, IscA from A. ferrooxidans has been studied. The results showed that IscA from A. ferrooxidans might function as a scaffold protein for the pre-assembly of Fe-S cluster and then transfer it to target proteins.
