Immobilization of Laccase on Modified Chitosan and Its Use for 2, 4-Dichlorophenol Degradation

The laccase was immobilized on crosslinking chitosan with glutaraldehyde and its properties for 2, 4-dichlorophenol degradation were evaluated. The activity of the immobilized laccase depending on the immobilizing procedure was discussed at length. The results showed that the activity of immobilized laccase prepared by crosslinking of chitosan and then adsorption was obviously higher than that by adsorption and subsequent crosslinking. For the crosslinking of chitosan, the optimum concentration of glutaraldehyde and crosslinking time was 5% and 12 h, respectively. For the adsorption of laccase, the immobilized laccase exhibited the maximum enzyme activity when laccase was adsorbed for 8 h at pH of 5.0. The optimum dosage of laccase was 90 mg/g carrier. The immobilized laccase showed obvious catalytic capacity for 2, 4-dichlorophenol degradation. When the amount of immobilized laccase was 8 g/L, removal efficiency of 2, 4-dichlorophenol in water (solution volume: 50 mL) reached 54.8% after 16 hours.
